摘要
目的建立小鼠局灶性脑缺血再灌注模型,并总结经验教训。方法从颈外动脉插入栓塞线至颈内动脉,选择性栓塞大脑中动脉,造成大脑中动脉闭塞的动物模型。通过控制再灌注时间,观察其对小鼠神经行为学、脑缺血程度及存活期的影响。结果除假手术组未制成脑缺血模型外,其他3组均于手术后24h出现神经行为学异常,脑切片TTC染色可见缺血病灶。结论小鼠局灶性脑缺血再灌注模型制造成功,小鼠术后的成活率、梗死体积、神经行为学表现与手术方法密切相关。
Objective In order to establish the focal cerebral ischemia reperfusion model in the mouse with suture method,and summarize experience from the experiment. Methods To establish a model of middle cerebral artery occlusion(MCAO) with introducing a suture into internal carotid artery(ICA) through external carotid artery(ECA), and occluding the middle cerebral artery(MCA). Through controlling the reperfusion time, observing the influence on neuroethology, extent of cerebral ischemia, survival time of the mouse. Results Except the sham operated group, other groups all emerged the change in neuroethology and ischemic focus in TTC after occluding 24 hours, Conclusion The model of focal cerebral ischemia reperfusion was successful, The survival rate, infart area and neumetholugy change of operated mouse were correlated with the method of operation.
