摘要
目的:检测大鼠肺纤维化时肺组织细胞凋亡及Fas/FasL基因的表达,探讨细胞凋亡在肺纤维化过程中的作用。方法:通过博来霉素复制大鼠肺纤维化模型,同时设立正常对照组,采用TUNEL法、RT-PCR和Western Blot法检测肺组织细胞凋亡、Fas/FasL mRNA以及蛋白的表达。结果:肺纤维化模型复制成功;模型组肺组织细胞凋亡率、Fas/FasL mRNA和蛋白表达均显著高于对照组。结论:细胞凋亡及Fas/FasL基因表达的上调在博来霉素致大鼠肺纤维化发生、发展的过程中起了重要作用。
Objective: To investigate the expression and significance of cell apoptosis and Fas/Fas-L gene in the lung tissue of rats with pulmonary fibrosis. Methods: The pulmonary fibrosis model in rat was established by bleomycin. TUNEL, RT-PCR and Western Blot were used to observe cell apoptosis and Fas/ Fas-L gene expression. Results: The model was reproduced successfully. Compared with the control group, cell apoptosis and Fas/Fas-L gene expression were significantly up-regulated in the rat's lung tissue in model group. Conclusion: The up regulation of expression of cell apoptosis and Fas/Fas-L gene may play an important role in the pathogenesis and development of pulmonary fibrosis.
