家蝇Attacin基因在家蝇三龄幼虫中的时间表达模式研究 被引量:6

摘要 目的探讨家蝇Attacin mRNA在幼虫各个时期的表达,为进一步研究家蝇的免疫防御打下基础。方法对家蝇三龄幼虫进行诱导,分别取诱导前0h和诱导后2h、4h、8h、12h、16h、24h、36h、48h的家蝇mRNA,进行RT-PCR,分析attacin基因在家蝇体内的时间表达模式。结果家蝇三龄幼虫体内的attacin基因在诱导前后均有表达,诱导后表达增强16h达到相对高水平,维持约24h,而后开始下降。结论Attacin基因在家蝇三龄幼虫中是组成性表达,对进一步了解其在家蝇免疫系统,丰富对昆虫天然免疫的认识具有积极作用。 Objective To explore the expression pattern of Attacin genes encoding in larva of Musca domestica for further studying of innate immunity. Methods The expression pattern of total RNA extracted from the unchallenged was analyzed and challenged third instar larva 2h, 4h, 8h, 12h, 16h, 24h, 36h, 48h after challenge respectively for determineating the level of Attacin gene by using semi - quantitative RT - PCR. Results The results demonstrated that transcripts of the Attacin gene were detected in challenged and unchallenged larvae, distinctly reduced at 16h, maintained to 24h , then decreased slowly. Conclusion Attacin gene has been expressed in the third instar larvae and it is of certain significance for further understand the immune system of Masea domestica.
