摘要
目的观察酒精性肝纤维化大鼠肝星状细胞(HSC)、肝细胞凋亡及清肝活血方对不同细胞凋亡的影响。方法将大鼠分为空白组,模型组,清肝活血低、中、高剂量组,易善复组。清肝活血方低剂量组〔4·75g/(kg·d)〕、中剂量组〔14·25g/(kg·d)〕、高剂量组〔28·5g/(kg·d)〕及易善复组〔66·5mg/(kg·d)〕,分别予相应药物灌胃,空白组和模型组以等量生理盐水代替,给药2周。比色法检测血清ALT、AST、γ-GT,HE、Masson染色观察肝脏炎症和纤维化程度。TUNEL法检测肝细胞凋亡,TUNEL-α-SMA双标记法检测HSC凋亡。结果清肝活血方能降低大鼠血清ALT、AST、γ-GT水平,减轻炎症及纤维化程度(P<0·05);诱导活化的HSC凋亡,减少肝细胞凋亡(P<0·05)。结论清肝活血方能有效减轻大鼠肝纤维化程度,并减少酒精引起的肝细胞凋亡,诱导活化的HSC凋亡。
Objective To observe the apoptosis of hepatic stellate cell (HSC) and hepatocyte and the effect of Qinggan Huoxue Recipe (QHR) on it in alcoholic liver fibrosis (ALF) rats. Methods Rats were divided into six groups: the normal group, the model group, the QHR low dose [4.75 g/(kg·d)], medium dose [14.25 g/(kg·d)] and high dose [28.5 g/(kg·d)] groups, and the Essentiale [66.5 mg/(kg·d)] group. They were treated with respective drugs through gavage for 2 weeks, while the normal and model groups were given normal saline instead. The serum levels of ALT, AST and γ-GT were measured by chromatometry, the degree of inflammation and fibrosis was observed by HE staining and Masson staining, and the apoptosis of hepatocyte and HSC were detected by TUNEL and TUNEL-α-SMA double immunolabeling respectively. Results Compared with those before treatment, the serum levels of ALT, AST and γ-GT obviously decreased ( P 〈 0.05), inflammation and fibrosis relieved ( P 〈 0.05), and the apoptosis of hepatocyte reduced ( P 〈 0.05 ), while that of activated HSC increased in the QHR groups after treatment (P 〈 0.05). Conclusion QHR can effectively relieve hepatic fibrosis, decrease hepatocyte apoptosis caused by alcohol and induce activated HSC apoptosis in ALF rats.
