摘要
目的:探讨雷公藤多甙(TWH)治疗类风湿关节炎(RA)的可能作用机制。方法:以佐剂性关节炎(AA)大鼠模型为研究对象(AA组),与经雷公藤多甙治疗后(TWH组)及正常对照组比较,每组12只。图像分析法检测关节软骨面积,免疫组织化学染色(SP)检测关节软骨中核因子κB受体激活剂(RANK)及核因子κB受体激活剂配体(RANKL)的表达。结果:与正常对照组大鼠相比,AA组大鼠滑膜增生,关节软骨破坏,软骨面积减少(P<0.05),软骨组织RANK及RANKL的表达增加(P<0.05);TWH组经过雷公藤多甙治疗后,与AA组相比,滑膜增生及关节软骨破坏减轻,软骨面积增加(P<0.05),软骨组织中RANK和RANKL表达量降低(P<0.05)。结论:雷公藤多甙可通过抑制滑膜增生及软骨组织表达RANK和RANKL而抑制AA大鼠软骨破坏。
Objective: To explore the possible role of glycoside of tripterygium wilfordii hook F (TWH) on rheumatoid arthritis (RA). Methods: Adjuvant induced arthritis rats (AA) was induced in Wistar rats. Arthritis rats were treated with TWH at the onset (14days) of arthritis. The area of articular cartilage was detected by image analysis. The expression of receptor activator of nuclear factor NF-κB (RANK)and receptor activator of nuclear factor NF-κB ligand (RANKL)in articular cartilage were detected by immunohistochemistry. Results. Compared with normal group, hyperplasia of synovial membrane was significant, the area of articular cartilage decreased (P 〈 0.05), and expression of RANK and RANKL in articular cartilage increased in AA control group (P 〈 0.05). By treated with TWH, hyperplasia of synovial membrane was depressed, the area of articular cartilage increased (P 〈 0.05), and the expression of RANK and RANKL in articular cartilage decreased in TWH group compared with that of AA group(P 〈 0.05). Conclusion: TWH may play a protective role in articular cartilage in RA by inhibiting hyperplasia of synovial membrane and expression of RANK and RANKL in articular cartilage.
