摘要
目的探讨KAI1基因和蛋白在正常胃黏膜、不典型增生胃黏膜及癌组织中的表达及其与之发生、发展的关系.方法应用原位杂交技术和免疫组织化学技术检测不典型增生胃黏膜65例及胃癌组织74例中KAI1 mRNA和KAI1蛋白的表达.结果KAI1 mRNA和KAI1蛋白在胃癌组织中表达显著降低,与正常胃黏膜、不典型增生胃黏膜组织中表达的差异有显著性意义(χ2=36.328,P<0.05).在胃癌组中不同的浸润深度、有无淋巴结转移和脉管侵犯组中阳性表达率的差异性有统计学意义(P<0.05);而在年龄、性别组间的差异性无统计学意义(P>0.05).结论KAI1 mRNA和蛋白低表达在胃癌组织中的发生、发展中可能起着重要作用,有望成为早期诊断、评估肿瘤细胞侵袭转移潜能的指标之一.
AIM: To study the expression of the metastasis suppressor gene KAI1 in gastric tumors and the relationship between the expression of KAI1 and the tumor genesis and development. METHODS: The expression of KAI1 mRNA and protein was investigated with in situ hybridization and immunohistochemistry (SP method) respectively in normal gastric tissues (n = 22), tumor tissues( n = 74) and tissues of atypical gastric mucosal hyperplasia(n = 65). RESULTS : The positive expression of KAI1 mRNA and protein in gastric carcinoma was significantly reduced and showed statistical difference as compared with normal gastric mucosa and atypical hyperplasia (x^2 = 36. 328, P 〈 0.05). The expression of KAI1 mRNA and protein in gastric carcinoma was correlated to the depth of infiltration of carcinoma, vessel invasion and lymph node metastasis (P 〉 0.05), but not to with patient's age and gender ( P 〉 0.05 ). CONCLUSION: KAI1 may play an important role in tumor genesis, development, invasion and metastasis of gastric carcinoma, and can serve as a molecular marker for early diagnosis and prediction of invasion and metastasis of gastric carcinoma.
