摘要
目的探讨小儿皮肤血管瘤各阶段肿瘤坏死因子相关凋亡诱导配体(TRAIL)的表达与细胞增殖和凋亡的关系。方法应用流式细胞术检测23例增生期、20例消退期血管瘤和15例正常皮肤组织中TRAIL蛋白的相对含量,细胞增殖指数(PI)和细胞凋亡率(AR)。结果增生期血管瘤、消退期血管瘤和正常皮肤组织中TRAIL蛋白的表达分别为1.26±0.56,1.74±0.68,1.00±0.23,PI分别为(40.98±12.02)%,(20.05±3.67)%,(15.64±4.01)%,AR分别为(11.96±4.02)%,(20.29±3.53)%,(6.61±1.29)%。3组间以上3指标两两比较,差异均有统计学意义。血管瘤组织中TRAIL蛋白的含量与AR呈正相关(r=0.36,P<0.05)。结论TRAIL蛋白诱导的凋亡可能参与了血管瘤由增生向消退的演变,细胞增殖和凋亡平衡关系的改变可能是血管瘤自行消退的重要原因之一。
Aim : To investigate the relationship between the expression of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL), cell proliferation and apoptosis in children's skin hemangioma. Methods:The level of TRAIL, the proliferation index (PI) and apoptosis rate (AR) were detected by flow cytometer (FCM) analysis in 23 case of hemangioma at proliferating phase, 20 cases of hemangioma at involuting phase, and 15 cases of normal skin. Results: In hemangioma at proliferating phase, hemangioma at involuting phase, and normal skin, the expressions of TRAIL were 1.26 ± 0.56, 1.74 ±0.68, and 1.00 ±0.23,PI was (40.98 ±2.02)% ,(20.05 ±3.67)% and (15.64 ±4.01)%, AR was (11.96 ± 4.02) %, ( 20.29 ± 3.53) % and ( 6.61 ± 1.29 ) %, respectively. There were significant differences among the 3 groups in the 3 parameters. The expression of TRAIL was positively correlated with AR ( r = 0.36, P 〈 0.05 ). Conclusion: TRAIL might be involved in the switch from uncontrolled growth to spontaneous involution of hemangioma, probably by inducing apoptosis. The equilibrium between cell proliferation and apoptosis might play a crucial role in the regression of hernangioma.
