摘要
目的探讨培养于诱导分化液中不同时间的大鼠神经干细胞体外诱导分化成多巴胺能神经元的效应。方法大鼠神经干细胞球在含有细胞因子IL-1、IL-11、GDNF和LIF的诱导分化液中分别培养2、4、6、8的10d后,流式细胞仪检测其体外诱导分化成多巴胺能神经元的分化率。结果在诱导分化液中大鼠神经干细胞球呈贴壁生长,球形结构开始塌陷,球内细胞从球体中央逐渐向四周分化扩展出较多形态各异的细胞,多数细胞有1、2个长突起或有多个短突起。免疫组织化学显示分化的细胞中含有TH阳性细胞,流式细胞仪检测诱导分化2、4、6、8和10d的神经球分化成TH阳性细胞比率分别为(3.2%±0.9%)、(6.8%±1.6%)、(16.7%±2.6%)、(14.8%±1.8%)和(12.2%±2.5%)。结论大鼠神经干细胞体外诱导分化成多巴胺能神经元存在着分化的时间效应,诱导分化6d的神经干细胞分化成多巴胺能神经元的比率最高。
[Objective] To investigate the effect on differentiation of rat neural stem cells which were cultivated in differentiate solution for different time to dopaminergic neurons in vitro. [Methods] We detected with flow cytometer the ratio of TH positive cells in differentiated cells derived from the neural stem ceils which were cultivated in differentiate solution containing IL-1α, IL-11, LIF and GDNF for 2, 4, 6, 8, 10 days respectively. [Results] In differentiate solution, the rat neural cell globes became attaching the dishs and began to collapse. Cells inside the globes grew out gradually and became irregular in shape. Most of the ceils had 1 to 2 long synapses or several short synapses. There were some TH positive ceils in the differentiated cells when detected with immunocytochemistry. The ratio of TH positive cells in differentiated ceils derived from the neural stem cells were (3.2%±0.9%), (6.8%± 1.6%), (16.7%±2.6%), (14.8%±1.8%), and (12.2%±2.5%) respectively. [Conclusion] There is time effect on differentiation of rat neural stem ceils to dopaminerglc neurons in vitro. The ratio of dopaminergic neurons in differentiat- ed ceils is the highest for neural stem cells which were differentiated for 6 days.
