摘要
对蝴蝶石斛兰高效再生体系的研究结果表明,将蝴蝶石斛兰杂交种子接种于3/4MS+BA 0.5 mg/L+NAA 1.0mg/L+活性炭1.0 g/L+白糖20.0 g/L+琼脂5.8 g/L培养基中培养60 d后可得到大量原球茎;将原球茎接种于MS+BA 2.0 mg/L+NAA 0.5 mg/L+椰子汁5%+白糖20.0 g/L+琼脂5.8 g/L培养基中培养45 d后大量增殖,60 d后可分化出小苗;将小苗接种于1/2MS+IBA 2.0 mg/L+NAA 1.5 mg/L+15%~20%椰子汁+白糖20.0 g/L+琼脂5.0 g/L培养基中培养,50 d后可形成具5~6片叶、2~4条根的健壮小苗;将小苗移植于混合基质(水苔或碎砖∶碎碳为41∶、蕨根和珍珠岩适量)中成活率达90%以上。
The high-efficient regeneration system of D.phalaenopsis. was studied in this paper.The results showed that lots of protocorms were induced when the seeds of D.phalaenopsis were cultured on 3/4MS medium supplemented with 0.5 mg/L BA, 1.0 mg/L NAA, 1.0 g/L active carbon, 20.0 g/L white sugar and 5.8 g/L agar for 60 days. The protocorms were multiplicated largely and were differentiated plantlets when cultured on MS medium supplemented with 2.0 mg/L BA, 0.5 mg/L NAA, 5% coconut water, 20.0 g/L white sugar and 5.8 g/L agar for 60 days.The plantlets developped to 5-6 leaves and 3-4 roots after cultured on 1/2MS medium supplemented with 2.0 mg/L IBA, 1.5 mg/L NAA, 15%-20% coconut water, 20.0 g/L white sugar and 5.0g/L agar for 50 days.And the survival rate was over 90% when the plantlets were transplanted to mixed stroma.
