摘要
分别研究了封闭液、血清稀释液及酶标二抗稀释液中BSA成分对牛结核抗体ELISA检测的影响。结果显示,酶标二抗稀释液中BSA的存在使ELISA检测值普遍偏低,且BSA的含量越高,检测值越低;为了提高ELISA检测值,使其达到理想检测效果,必须提高酶标二抗的使用浓度。BSA中可能存在的牛血清抗体杂质对酶标二抗起到了“中和”作用,降低了酶标二抗与目标抗体结合的实际浓度,使检测结果普遍偏低。
In this study, bovine serum albumin(BSA) was used as components of blocking buffer, diluents for serum and enzyme-labeled secondary antibody, respectively. And the effects of BSA on ELISA which was used to detect antibodies of Mycobacterium boris was discussed. The component of BSA in diluent for enzyme-labeled secondary antibody was found to significantly lower the read of OD630 in ELISA test, and with the increasing of BSA in diluent for enzyme-labeled secondary antibody, the read of OD630 in ELISA test fell. In order to heighten the read of ELISA test and reach an ideal detection effects, we had to heighten concentration enzyme-labeled secondary antibody. Based on the above research, we got that there was some antibody of bovine serum in BSA. The enzyme-labeled secondary antibody was counteracted by these antibody,and the concentration of which was reduced when it reacted with target antibody. Then read of ELISA test was lowered.
