摘要
为了筛选出鉴定小麦背景下黑麦成分的通用特异性引物,选用胜利黑麦、八倍体小黑麦新麦73、六倍体小黑麦哈师209、普通小麦s165、中国春为试验材料,利用国内外发表的黑麦特异性DNA探针引物进行PCR扩增并进行了电泳检测。结果表明,在选用的6对引物(NOR-R1、AF1/AF4、PASW、pSC19.1、pSC20H、pSC10C)中只有4对引物即pSC20H、pSC119.1、AF1/AF4、NOR-1R在黑麦和小黑麦DNA中扩增出了特异性条带,扩增片段大小依次为1490、750、1500、300 bp左右,说明这4对引物是黑麦通用特异性DNA探针引物,可以用于在小麦背景下进行黑麦外源种质的鉴定。
With the introgression of good alien genomes, it is important to detect the segment of alien chromatin in wheat background. In order to screen the universal specific DNA probes of rye, using rye, triticale, wheat were used as experiment material, PCR with universal primers of rye were conducted and PCR products were examined electrophoresis. Only 4 pairs of primers could produce specific band among 6 pairs of candidate primers and could be used as the universal specific DNA probes of rye to identify rye alien chromatin in wheat background.
