椎间盘髓核细胞中Sox9与Ⅱ型胶原基因表达的关系 被引量:7

摘要 目的:探讨椎间盘髓核组织中Sox9基因表达的变化及其与Ⅱ型胶原基因表达的关系。方法:将30个椎间盘组织按Thompson分期分为Ⅰ~Ⅳ期,应用RT-PCR、Western blot和免疫组化方法检测各期间盘组织中Sox9、Ⅱ型胶原基因的mRNA和蛋白表达;应用SPSS10.0统计学软件,采用单因素方差分析、t检验和Pearson相关性检验分析两者的相互关系。结果:椎间盘髓核组织中Sox9mRNA的表达量在总体上低于Ⅱ型胶原,Sox9蛋白表达位于细胞核中而Ⅱ型胶原主要位于细胞间质内,Sox9在ThompsonⅠ期椎间盘的细胞核内表达很强而在Ⅳ期则很弱甚至缺失;从ThompsonⅠ~Ⅳ期两种基因的mRNA和蛋白表达水平均逐渐降低,各分期间有显著性差异(P<0.05)。ThompsonⅠ~Ⅳ期标本中Ⅱ型胶原与Sox9表达量的下降趋势相近。结论:Sox9和Ⅱ型胶原基因表达水平与Thompson分期密切相关,随椎间盘退变程度加重表达逐渐降低,且两者下降趋势相近。 Objective:To investigate gene regulation meehanism in the intervertebral disc by studing the correlation between the expression of Sox9 gene and collagen type Ⅱ gene,Method:30 human intervertebral discs taken from surgery were classified into four grades according to Thompson grading system.The expression of Sox9 and collagen type Ⅱ genes were detected with reverse transcriptional PCR (RT-PCR),Western blot and immunohistochemistry,and correlative extent between the two markers was also evaluated.By SPSS 10.0, ANOVA,t test and Person correlation test were chosen to test the results.Result:The expression of Sox9 mRNA in the total amount was less than that of collagen type Ⅱ,Sox9 was seen localized in NP cell nucleus while collagen type Ⅱ localized in the matrix.Sox9 had a high expression in the cells of the Thompson grade Ⅰ nucleus pulposus,but had a weak or absent expression in cells of Thompson grade Ⅳ,The expression in both mRNA and protein level for the two markers showed a steping down tendency in the samples from Thompson Ⅰto Thompson Ⅳ(P〈0.05).Pairwise analysis showed a similar tendency between the expression of Sox9 and collagen type Ⅱ .Conclusion:The expression of Sox9 and collagen typeⅡ in the NP cell of human discs is associated with the Thompson grade closely,and the more severe the degeneration of the disc,the closer the tendency of the two matters.
