转基因草莓植株cNHX1基因表达水平和拷贝数的Real Time PCR分析 被引量:2

摘要 分析了转cNHX1基因的草莓不同植株的外源基因表达水平和cNHX1基因的拷贝数。结果表明:不同转基因株系cNHX1基因表达水平存在差异,而且这种差异随盐胁迫时间的延长而更明显。在6个株系中,T6株系有最高和最稳定的cNHX1基因表达水平,而T2株系的表达水平最低。对转基因植株基因组cNHX1基因拷贝数的分析表明,T3、T6、T8和T14的含量相似,而与T2株系相差289%~310%,推测T3、T6、T8和T14株系为单拷贝植株,而T2株系为3拷贝株系。 The expression level of cNHX1 gene of different transgenic strawberry lines were analyzed by using Real Time PCR, the results manifested the existence of difference in expression level among these lines, and this difference was more notable followed with the prolonged NaC1 treating time. Among these lines, T6 lines had the most higher and steady expression level, while T2 lines had the most lower expression level. The relative cNHX1 fragment contents in genome of transgenic lines were analyzed also in this experiment, the results showed T3,T6,T8 and T14 lines had similar contents, and apparently different with it in T2 line(variety from 289% to 310% ,respectively). By compared with the preliminary data of the genome size of strawberry, we speculated that T3,T6,T8 and T14 lines were one copy inserted lines, and T2 was 3 copy inserted line.
