摘要
以明胶作为载体,采用包埋技术成功地将鸡白痢抗体修饰于电极表面,制成非标记型鸡白痢免疫传感器。采用循环伏安法和交流阻抗图谱对不同修饰状态的电极进行表征。根据抗原抗体特异性结合形成的免疫复合物使敏感膜有效扩散截面积减小的特性,采用循环伏安法以铁氰化钾为分子探针间接检测鸡白痢抗原。通过试验确定鸡白痢阴阳性判定标准,Δi>2.5μA判为阳性,Δi<2.5μA判为阴性。该免疫传感器特异性强,重现性好(RSD<6%),准确性高(与国标检测方法符合率为88.9%),4℃干态放置7d,其响应电流为初始电流的90.2%。因此该免疫传感器可初步用于鸡白痢阴阳性判定和快速筛选。
Salmonella pullorum antibody was successfully modified on the platinum electrode surface to fabricate the label-free immunosensor by embed technology with gelatin as matrix. In the experiment, the electrochemical character of the immunosensor was characterized by cyclic voltammetry(CV)and electrochemical impedance spectroscopy(EIS). Cyclic voltammetry could be used to indirectly detect Salmonella pullorum antigen with kalium ferricyanide as molecule probe because the effective diffusion section area has been decreased along with immunocomplex increasing caused by antigen-antibody immunoreaction, The positive and negative determinant standards confirmed by experiment are as follows: △i〉2.5μA is positive, △i〉2.5μA is negative. Specificity, reproducibility(RSD〈6%) and accuracy(the results obtained by immunosensor 80% according with GB/T 4789.4-2003) were studied, and its response current reached 90.2% of its initial current when stored in dry air at 4℃ for 7 days. Therefore, the immunosensor could be used to practically detect Salmonella pullorum.
