摘要 目的探讨葛根素对成年大鼠创伤性脑损伤神经细胞中Bcl-2、Bax蛋白表达的影响。方法不同浓度葛根素作用于创伤性脑损伤神经细胞,通过病理学检查、Tunel法及免疫组织化学法检测神经细胞凋亡及Bcl-2、Bax蛋白的表达。结果葛根素作用脑损伤神经细胞7天后,以浓度效应关系改善创伤性脑损伤神经细胞的病理改变,减轻细胞凋亡程度,增加Bcl-2的表达,降低Bax/Bcl-2比率,但不影响Bax的表达。结论葛根素对创伤性脑损伤神经细胞具有保护作用,且呈剂量依赖性,其机制可能与葛根素抗细胞凋亡,增加Bcl-2的表达有关。 Objective To investigate the effect of Puerarin on the expression of Bcl-2 and Bax protein of neural cells following traumatic brain injury (TBI) in adult rat. Methods After treatment by different concentrations of Puerarin on neural cells following traumatic brain injury, the effect of Puerarin on the expression of Bcl-2 and Bax proteinum was investigated with pathologic observation,apoptosis of neurocyte as mediated by dUTP biotin bick end labeling (TUNEL) and immunohistochemical method. Results After Puerarin treatment for 7 days,the level of pathologic alteration of neural cells was improved in concentration-dependent manner,and mitigated apoptosis was improved,the number of Bcl-2 positive cells increased and the proportionality of Bax/Bcl-2 depressed. But the expression of Bax proteinum was not interfered by Puerarin. Conclusion The results showed that Puerarin preserved neural cells in the adult rat following traumatic brain injuy (TBI) in concentration-dependent manner. The mechanism of which may be related with anti-apoptosis effect and the increased expression of Bcl-2 protein.
