调心方有效部位TX0201对类AD模型大鼠学习记忆和细胞凋亡相关基因的影响 被引量:12

摘要 目的:观察调心方有效部位TX0201对Aβ1-40双侧杏仁核注射所致的类阿尔茨海默病(AD)大鼠模型学习记忆和皮层、海马细胞凋亡相关基因的影响。方法:应用β-淀粉样蛋白1-40片段(Aβ1-40)注射入大鼠双侧杏仁核建立类AD的动物模型;实验设正常对照组、类AD模型组、TX0201组、调心方组、阳性药安理申组,均采用灌胃给药20 d;用Morris水迷宫法测试大鼠学习记忆能力;RT-PCR法检测大鼠脑部皮层和海马的细胞凋亡相关基因Bax mRNA,Bcl-2 mRNA。结果:TX0201、调心方和安理申均能够改善类AD大鼠的学习记忆能力,调心方能全面改善模型鼠脑部皮层和海马异常表达的Bax mRNA和Bcl-2 mRNA,TX0201可改善模型鼠皮层和海马异常表达的BaxmRNA,安理申组改善模型鼠皮层异常表达的Bax mRNA。结论:TX0201可通过改善模型鼠脑内的Bax mRNA来抑制细胞凋亡,从而改善模型鼠的学习记忆。 Objective: To research the mechanism of TX0201 abstracted from heart-regulating formula in the treatment of rat with Alzheimer's disease (AD). Method: The rat models of AD were induced by Aβ1-40 injected into the bilateral amygdale. All rats were divided into five groups at random, namely control group, model group, heart-regulating formula group, TX0201 group and aricept group. The rats were intragastricaily treated with different solution respectively for 20 days. The effect of TX0201 on spatial learning and memory ability of these rat models was investigated with the method of Morris water maze. By using the method of RT-PCR, the expression of apoptesis associated genes (such as Bcl-2 and Bax) in cerebral cortex and hippocampus of rats were examined. Result: Heart-regulating formula, TX0201 and aricept could significantly improve the spatial learning and memory ability of rats. Heart-regula- ting formula could comprehensively redress the abnormal expression of Bax mRNA and Bcl-2 mRNA in cerebral cortex and hippocampus. TX0201 could rectify the abnormal expression of Bax mRNA in cerebral cortex and hippocampus. Aricept could regulate the abnormal expression of Bax mRNA in cerebral cortex. Conclusion: TX0201 could ameliorate the learning and memory ability of these model rats by alleviating neuron apoptosis by means of regulating the abnormal expression of Bax mRNA in brain.
