摘要
目的:建立油含量的化学检测方法,并对聚醚类抗生素发酵液中的残油含量进行检测。方法:油水解为脂肪酸后与Cu^2+离子反应生成蓝色的油酸铜,用比色法检测油酸铜的量,并计算发酵液中残油的量。检测波长为715nm。结果:该方法的线性范围0.4-2.0mg/ml,回归系数为R^2=0.9999,平均RSD为0.466%,回收率在96.88-102.33%之间,以油和水制成的指示管,平均相对误差为-0.17%。结论:该方法线性、重复性和回收率良好,检测灵敏度高,可作为聚醚类抗生素发酵过程中残油指标的检测。
Objective: To establish a colorimetry for the determination of oil in the broth of the polyether antibiotics fermentation.Method: The oleic acid which the oil was hydrolyzed into react with Cu^2+,and the cupric oleate was found.Calculation of oil content was performed by detection of the cupric oleate content at 715nm.Results:The calibrated linear curve was in the range of 0.4~2.0mg·ml^-1(R^2=0.9999),RSD was less than 0.50%,recovery rate was 96.88~102.33%,average relatively error of five test is-0.17%.Conculsion:The linear ,reproducible and sensitive were confirmed.
