摘要
从小麦粉中提取植物酯酶,用70%饱和硫酸铵对植物酯酶进行纯化,酶催化反应温度在45℃、pH4.5~5.5时酶活性最大,酶催化反应动力学显示10min内反应呈线性关系。利用有机磷农药能抑制植物酯酶酶促反应的机理,建立了快速测定粮食中有机磷农药残留的方法。测定有机磷农药残留反应体系最大吸收峰是在480nm之间,最适乙酸-β-萘酯用量为1.0mL;在农药浓度在0.1~1.0mg/kg之间时,所到之处得到的吸光值与农药浓度呈线性相关;在青菜汁、茶叶汁和大米提取液回收率分别为89%、84%和93%。
The plant - esterase extracted from wheat flour was purified by 70% saturated ammonium sulfate. The obtained plant -esterase exhibits its maximum activity at 45℃ and pH 4.5 - 5.5, and exhibits a line relation of reactive velocity vs reactive time within 10 min. A rapid detection method of organophosphorus pesticide residues was developed on the following principle : the organophosphorus pesticide can inhibit the plant - esterase activity. The reaction system of the organophosphorus pesticide determination shows the following characters : maximum peak on 480 nm, optimal dosage of acetic-β -naphathone 1.0 mL, line relation of absorption value vs organophosphorus pesticide concentration in the pesticide concentration range of 0.10 - 1.0 mg/kg, recovery rate of 89%, 84%, and 93% for China vegetable juice, tea juice, and extract of rice, respectively.
