摘要
目的探讨毛细管区带电泳分离大肠埃希菌的有关影响因素。方法用毛细管区带电泳法对大肠埃希菌ATC-C25922进行了分离研究,分析缓冲液pH值、添加剂PEO分子量及浓度、电压和毛细管长度对分离的影响。结果缓冲液pH值、添加剂PEO分子量及浓度、电压和毛细管长度对毛细管区带电泳分离大肠埃希菌有较大影响,在优化的条件下,大肠埃希菌ATCC25922能达到很好的分离。结论调节贮存缓冲液TBE(4.5 mmoL/L Tris,4.5 mmoL/L硼酸,0.1mmoL/L EDTA)pH值至9.0,添加剂0.0125 g/dl PEO(Mr600 000),毛细管总长度50.2 cm,电压25 KV,毛细管区带电泳分离大肠埃希菌ATCC25922,分离时间20 min,峰形尖锐,效果最佳。
Objective To investigate some influential factors for determination of Escherichia coli ATCC25922 by capillary zone electrophoresis. Methods Escherichia coli ATCC25922 were determined by capillary zone electrophoresis. Buffer pH, additives PEO molecular weight and concentration,voltage,and capillary length were studied and optimized for fast and efficient separation. Results Buffer pH, additives PEO molecular weight and concentration, voltage,and capillary length could improve the separation profile,Escherichia coli ATCC25922 were well separated on best condition. Conclusion The stock buffer(4. 5 mmoL/L Tris,4.5 mmoL/L boric acid,and 0.1 mmoL/L EDTA)was adjusted to pH9.0,additives 0. 0125 g/dl PEO (Mr 600 000),capillary length 50. 2 cm,voltage 25 KV,migrating 20min,determining Escherichia coli ATCC25922 by capillary zone electrophoresis showed the peak focused into sharp and achieved extremely high efficiencies.
