Study advance of human telomerase reverse transcriptase

摘要 人端粒酶是染色体末端的DNA蛋白酶,对维持染色体稳定至关重要,在细胞增殖、衰老、永生化及癌变中起重要作用。人端粒酶逆转录酶(hTERT)是端粒酶激活的主要限制因素,与肿瘤的发生发展密切相关,因此,认识其调控机制及在肿瘤诊治的研究广泛开展。目前,hTERT用于临床肿瘤诊断的意义已被认可,并且已有利用其进行肿瘤治疗的Ⅰ期临床试验。 The human telomerase is a DNA-binding protease at the end of the chromosome,which is essential to maintainning telomere stability, and it has important effect to proliferation, senility, immortalization and canceration of human cell. The human telomerase reverse transcriptase(hTERT) is a main limit factor of the activation of telomerase,which closely correlates with tumorous genesis and growth. Therefore,studies of the hTERT regulative mechanisms and in the tumorous diagnosis and therapy were extensively evolved. At present,hTERT used in clinical diagnosis of tumor was approved and was used in therapy tumor in stage Ⅰ clinical trial.
