白细胞介素-1β在去卵巢大鼠骨髓基质细胞中的表达

摘要 目的:观察卵巢切除后大鼠胫骨形态计量学改变及体外培养的骨髓基质细胞(BMSC)中白细胞介素-1β(IL-1)βmRNA表达。方法:将24只4月龄雌性Wistar大鼠随机分为去卵巢1、4个月组,假切1、4个月组,分别于实验末断颈处死,取左侧胫骨,新鲜分离骨髓细胞,静置贴壁法体外培养13d后,采用原位杂交法检测BMSC中IL-1βmRNA表达。取右侧胫骨制备不脱钙的骨切片,应用目镜网格观察胫骨形态计量学参数改变。结果:与假切1个月组相比,去卵巢1个月组大鼠胫骨成骨表面(Ob.S/BS)、类骨质表面(OS/BS)显著减少(P<0.01,P<0.05),吸收表面(ES/BS)、破骨细胞数(N.Oc/T.Ar)显著增加(P<0.05);与假切4个月组相比,去卵巢4个月组胫骨平均骨小梁体积(BV/TV)显著减少(P<0.05),ES/BS显著增加(P<0.01),N.Oc/T.Ar显著增多(P<0.05)。在体外培养的BMSC原位杂交中,IL-1βmRNA均能在扁圆形细胞和纺锤形细胞中表达,阳性细胞的胞浆呈棕黄色,其中在扁圆形细胞中的表达较强,去卵巢1、4个月后IL-1βmRNA在骨髓基质扁圆形细胞的表达明显高于对照组(P<0.01,P<0.05)。结论:去卵巢后,随时间推移大鼠胫骨骨质丢失严重,IL-1βmRNA在体外培养骨髓基质扁圆形细胞中的表达明显增强。 Objective:To observe the expression of interleukin-1β (IL-1β) mRNA on cultured bone marrow stromal cells (BMSC)and the change of tibia histormorpbometry of ovariectomized rats.Methods:Twenty four female Wistar rats (4-month old)were randomly divided into 4 groups:ovariectomy for 1 and 4 months,and sham-ovariectomy for 1 and 4 months.All rats were sacrificed by neck dislocation at the end of experiment time.The BMSC were freshly isolated from bone marrow of left tibia,then cultured by static adherent culture for 13 days,and their right proximal tibia were processed undecalcified sections.The index of tibia histormorphometry was measured with grid.The expression of IL-1mRNA on cultured BMSC were investigated by means of in situ hybridization.Results:By the end of first month after operation,bone forming index including Ob.S/BS and OS/BS decreased(P〈0.01,P〈0.05),but bone resorption index including ES/BS and N.Oc/T.Ar increased (P〈0.05).After forth month, BV/TV decreased (P〈0.05), and bone resorption increased (P〈0.05, P〈0.01 ). Both spindle-like and flat-like BMSC expressed IL-1βmRNA.Compared with the control group,the expression of IL-1βmRNA on Cultured flat-like BMSC increased in the experimental group (P〈0.05).Conclusion:As time goes on,expression of IL-1βmRNA on cultured fat-like BMSC increases and bone loss in tibia is obvious after ovariectomy.
