Increased expression of NADPH oxidase p47^(-PHOX) and p67^(-PHOX) factor in idiopathic pulmonary fibrosis

摘要 目的探讨特发性肺纤维化(IPF)患者支气管肺泡灌洗液(BALF)和外周血中性粒细胞膜蛋白质和还原型辅酶Ⅱ(NADPH)氧化酶蛋白亚单位 p47^(-PHOX)和 p67^(-PHOX)因子磷酸化表达。方法选择15例 IPF 患者为病例组,7例无器质性肺疾病者为对照组。采用细胞色素 C 还原法测定外周血中性粒细胞释放超氧阴离子水平,采用免疫细胞化学法和 Western 免疫印迹法检测 BALF 和外周血中性粒细胞膜蛋白质和 NADPH 氧化酶 p47^(-PHOX)和 p67^(-PHOX)因子磷酸化表达。结果 BALF 中性粒细胞膜蛋白质磷酸化,IPF 组平均吸光度值(0.19±0.02)显著高于对照组(0.14±0.01);IPF 组外周血中性粒细胞膜相对分子质量为80 000、58 000、45 000蛋白质的磷酸化表达和 p47^(-PHOX)和 p67^(-PHOX)因子的磷酸化表达(强阳性)均较对照组(弱阳性)显著增强。IPF 组中性粒细胞释放超氧阴离子的水平与用力肺活量、第一秒用力呼气容积、动脉血氧分压、一氧化碳弥散量呈显著负相关。结论外周血中性粒细胞释放超氧阴离子水平与肺纤维化程度存在一定关系。NADPH 氧化酶 p47^(-PHOX)和 p67^(-PHOX)因子在 IPF 患者中表达增加。 Objective To investigate the phosphorylation of the proteins, NADPH oxidase p47^-PHOX and p67^-PHOX factor in neutrophil cell membranes from bronchoalveolar lavage fluid (BALF) and peripheral blood of patients with idiopathic interstitial fibrosis ( IPF ). Methods Fifteen patients with IPF ( IPF group) and 7 controls without lung diseases (control group ) were included. The superoxide anion of peripheral blood neutrophils were measured using the cytochrome C reducing method. Immunocytochemical method and Western blotting were used to measure the phosphorylation expression of cell protein in BALF and phosphorylation of membrane protein and p47^-PHOX and p6T^-PHOX factor in peripheral blood neutrophils, respectively. Results The phesphorylation of BALF neutrophils ( average absorbance) was 0. 14 ± 0. 01 in the control group, 0. 19 ± 0. 02 in the IPF group, the difference being significant. The phosphorylation levels of relative molecular weight 80 000, 58 000, and 45 000 membrane proteins and p47^-PHOX and p67^-PHOX of the peripheral blood neutrophils in the IPF group were higher than those in the control group. The level of superoxide anion in the peripheral blood neutrophils of the IPF group was negatively correlated with forced vital capacity, one second forced expiratory volume, PaO2, diffusing capacity of the lung for carbon monoxide. Conclusion The level of superoxide anion of the blood neutrophils is associated with pulmonary fibrosis. There is an increased expression of NADPH oxidase p47^-PHOX and p67^-PHOX factor in IPF patients.
