摘要
采用自行建立的圆盘等电聚焦电泳将大肠组织中芳香基酰胺酶(AAD)分离出10条区带,从阳极到阴极依次为AADⅠ~AADⅩ,其中AADⅥ主要见于大肠癌组织(75%)、癌旁组织(50%),面远癌组织仅9.1%,前二者与后者有显著性差异(p<0.05)。大肠癌中AADⅥ与Dukes分期和分化程度无相关性(p>0.05),其等电点和胎盘型AAD(P-AADⅡ)等电点一致(PI6.33)。结果表明,AADⅥ是大肠癌组织异位表达的证据,可望成为大肠癌及癌前病变的标志物。
By means of cylindrical isoelectric focusing,Large bowel tissue arylamidase was separated into ten isoforms, which were named AADⅠ-AADⅩ from anode to cathode,AADⅥ was mainly present in large bowel carcinoma(75.0%)and the tissue adjacent to cancer(50.0%),the tissue remote to cancer (only 9.1%)(P<0.05).There was no correlation among AADⅥ,dukes period and cell classification (p>0.05),AADⅥ was identical with placental type AAD(P-AADⅡ)in isoelectric point (PI 6.33). Clinical research showed that AADⅥ was evident of ectoexpression in large bowel carcinoma tissue and that might be regarded as biochemical marker for diagnosing cancer and precancerous lesions.
