摘要
为探讨牙鲆生长的分子遗传机理,选取同一饲养环境下生长的60日龄牙鲆,根据体重和体长的显著差异分成大小两组,筛选出14对引物组合,应用mRNA差异显示技术,研究了两组牙鲆肌肉组织的基因表达差异。在8%的聚丙烯酰胺凝胶上共获得241条可重复扩增的条带,占可扩增条带(288条)的77.43%。其中有223条为共有条带,占总条带数(241)的92.53%;差异表达的片段有18条,占总条带数(241)的7.47%;它们只表现为强弱的差异,没有发现“有或无”的基因表达差异现象。实验结果说明:在长期相同的人工养殖条件下,牙鲆肌肉组织中基因的mRNA表达差异较少;相同养殖环境条件下表现出不同生长速度的牙鲆鱼肌肉组织中所存在的强与弱的基因表达差异的原因,可能源于调控其肌肉组织生长发育基因的内含子调控序列存在某些差异。
In 9rder to understand molecular genetic basis of paralichthys olivaceus growth, mRNA differential display was used to analysis the differences in gene expression of musculature between two groups which 60 days old classfied with weigh and length. 14 primer combination was used and 241 repeated bands were obtained from 8% PAGE in which included 223 mutual bands about 92.53% of it. It was about 77.43% of total bands(288) and Only one pattern of gene expression was observed. Our results indicate that there are little difference between musculature of paralichthys olivaceus feed in the same environment and difference expression between two groups maybe origin from regulated sequence of some introns which could be closely related to growth speed.
