摘要
目的探讨大鼠视神经损伤后上丘BMP4 mRNA与Noggin mRNA的表达变化。方法采用单侧眼球摘除的视神经损伤动物模型,将大鼠分成正常对照组、眼球摘除后24 h、1周、2周、3周组,原位杂交检测对侧上丘BMP4 mRNA与Noggin mRNA的表达变化,免疫组化检测MAP2和GFAP阳性细胞数量和形态的变化。结果BMP4 mRNA在损伤后24 h显著升高(P<0.01),1周组的表达较24 h降低;Noggin mRNA在损伤后各组的表达水平与正常对照组相比无明显变化(P>0.05)。GFAP阳性细胞在损伤后24 h即明显增加(P<0.01),MAP2阳性细胞在损伤后1周数量减少(P<0.01)。结论单侧眼球摘除后大鼠对侧上丘BMP4 mRNA与Noggin mRNA的表达水平可能是导致损伤后星形胶质细胞反应性增生和神经元退行性死亡的因素之一。
Objective To investigate the changes of BMP4 mRNA and Noggin mRNA in superior colliculus after optic nerve injury. Methods Unilateral eyeball enucleation was used as animal model of optic nerve injury. Wistar rats were randomly divided into control group, and 4 groups including 24 h, 1 week, 2 weeks and 3 weeks after injury. The expression of BMP4 mRNA and Noggin mRNA in contralateral superior colliculus was detected by in situ hybridization. GFAP and MAP2 was detected by immunohistochemical method. Results BMP4 mRNA massively increased in 24 h after injury (P 〈0. 01 ) and decreased slightly in 1 week. Noggin mRNA had no obvious changes in the four groups as compared with control group (P 〉 0. 05). GFAP IR cells increased in 24 h after injury (P 〈 0.01 ) ; MAP2 IR cells decreased since 1 week after injury ( P 〈 0. 01 ). Conclusion The expressions of BMP4 mRNA and Noggin mRNA in contralateral superior colliculus after unilateral eyeball enucleation may be one reason of the increase of astrocytes and degeneration of neurons.
