摘要
利用RAPD和ISSR分子标记技术对11份云南核桃(Juglans silillata Dode)品种进行了遗传多样性研究。研究结果表明,筛选出的8条RAPD和8条ISSR引物分别产生86和102条扩增带,其中多态性条带分别为53和62条,分别占总数的61.63%和60.78%。用POPGENE对两种标记方法的遗传多样性进行计算,有效等位基因数分别为1.3552和1.3707,基因多样性为0.2110和0.2143,Shannon信息指数为0.318 8和0.3216。以Nei氏遗传距离矩阵按UPGMA方法聚类分析结果RAPD和ISSR标记的遗传距离分别为0.0355~0.4113和0.1423~0.4011,平均值各为0.2288和0.2338,以上数据表明分析品种具有比较丰富的遗传变异,同时也说明尽管Mantel相关性检测两种标记方法相关性较低(r=0.543 9,P〈0.05),但都可以有效地揭示核桃品种的遗传多样性状况。
The genetic diversity of 11 Juglans silillata Dode varieties in Yunnan have been investigated with the techniques of RAPD and ISSR.The results show that 86 and 102 bands were obtained by RAPD and ISSR markers amplified through 8 selected primers respectively.53 and 62 are polymorphic,which account for 61.63% and 60.78% respectively.Analyzed through POPGENE,effective number of alleles per locus,gene diversity and Shannon's information index are 1.355 2,0.211 0 and 0.318 8 by RAPD,and those of by ISSR were 1.370 7,0.214 3 and 0.321 6 respeetively.The UPGMA clusteting indicated that the genetic distance was 0.035 5 -0.411 3 by RAPD and 0.142 3-0.401 1 by ISSR,of which the average value was 0.228 8 and 0.233 8 respectively. The above results show that there is a relatively rich genetic variation in the varieties tested and that both techniques are effective in revealing genetic diversity in Juglans silillata Dede although they had a lower correlation (r=0.543 9, P 〈0.05).
