感染登革2型病毒的白纹伊蚊卵在(25±1)℃、光照14h/天,75%±5%RH的相对干燥环境中保存42天,分别采用C6/36细胞培养分离病毒和RT-PCR方法,检测卵孵化的F1代蚊虫感染率。第1个生殖营养周环F1代蚊虫未检测和分离到病毒,PCR检测第2生殖营养周环蚊虫批阳性率为26.7%,最低感染率为1∶112.5;第3生殖营养周环蚊虫批阳性率为27.8%,最低感染率为1∶108。统计学检验结果表明第2和第3生殖营养周环卵孵化的F1代蚊虫感染率没有达到显著性水平(P>0.05)。结果表明感染白纹伊蚊卵在75%±5%RH的相对干燥环境中保存42天后,能在孵化的子代蚊虫中检测和分离到病毒,证实伊蚊卵在干燥环境中能够保存卵内病毒。
Whether dengne-2 virus (DENV-2) can survive in infected desiccated eggs of Aedes albopictus (Skuse) was determined in the study. Eggs of Ae. albopictus infected with DENV-2 were placed into a desiccation environment at (25 ± 1 )℃, and a daily photoperiod of 14 h light with (75 ± 5 ) % RH for 42 d. C6/36 cells and reverse transcriptlon-polymerase chain reaction (RT-PCR) were used to detect virus in F1 progeny derived from desiccated eggs. Three gonotrophic cycles (GC) eggs were collected. Vertical transmission was not observed in the first gonotrophic cycle. The total positive rate of GC2 progeny was 26.7 %, the minimum infection rate was 1: 112.5. As for the GC3 progeny, there were 27.8% and 1 : 108, respectively. There was no significant difference observed between the infection rates of GC2 and GC3 progeny ( P 〉 0.05). The results indicated that DENV-2 can survive in desiccated eggs of infected Ae. albopictus and also be detected in F1 progeny.
