摘要
目的:观察佐剂关节炎(adjuvant arthritis,AA)大鼠肺功能、细胞因子含量的变化及新风胶囊对其影响。方法:将60只大鼠随机分为正常组、模型组、甲氨喋呤(MTX)组、雷公藤多苷片(TPT)组和新风胶囊(XFC)组,每组12只,分别向除正常组以外的其余动物右后足跖皮内注射弗氏完全佐剂0.1 ml致炎,致炎后第19天开始给药。正常组及模型组均给予9.0 g/L氯化钠注射液,其余3组分别给予MTX、TPT、XFC。观察各组大鼠肺功能及血清细胞因子的变化。结果:与正常组比较,模型组及MTX组的用力肺活量(FVC)、第一秒用力呼气容积(FEV1)、25%肺活量的最大呼气流量(FEF25)、50%肺活量的最大呼气流量(FEF50)、最大呼气中段流量(MMEF)、最大呼气流量(PEF)显著降低(P<0.05,P<0.01),TPT组的FVC、PEF显著降低(P<0.05);与模型组比较,XFC组的FVC、FEV1、FEF25、FEF50、MMEF、PEF显著升高;与XFC组比较,MTX组的FVC、FEV1、FEF25、FEF50、MMEF、PEF显著降低(P<0.05,P<0.01),TPT组的FVC、FEF50、MMEF显著降低(P<0.05)。与正常组比较,模型组TNFα显著升高(P<0.05);与模型组比较,XFC组TNFα显著降低(P<0.01),而与其余两个治疗组无显著差异(P>0.05);与正常组比较,模型组IL-10显著降低(P<0.01);与模型组比较,XFC组IL-10显著升高(P<0.01),而与其余两个治疗组无显著差异(P>0.05)。结论:佐剂关节炎大鼠存在通气功能及小气道功能障碍,新风胶囊能改善AA大鼠肺功能,其机制可能与新风胶囊下调致炎因子(TNFα)、上调抗炎因子(IL-10)、抑制致炎效应、增强抗炎效应有关。
Objective : To observe the changes of pulmonary function and cytokines (TNFα, IL-10) in serum of adjuvant arthritis rats and influences of Xinfeng capsule (XFC) on these changes. Methods: Sixty rats were averagely divided into normal group, model group, Methotrexate (MTX) treated group, tripterygium glycosides tablet (TPT) treated group, XFC treated group, 12 in each. Except for the rats of normal group, the others were intracutaneously injected with 0. 1 ml of Freund's complete adjuvant in the right hindlimb. After 19 days, different drugs were given to all rats. The pulmonary function was determined and the blood samples were reserved for assaying cytokines. Results: The forced vital capacity (FVC), forced expiratory volume in first 1 second( FEV1 ) ,maximal expiratory flow at 25 percentage of forced vital capacity ( FEF25 ), maximal expiratory flow at 50 percentage of forced vital capacity( FEF50) ,maximal mid-expiratory flow rate(MMEF) and peak expiratory flow(PEF) of model group and MTX treated group were obviouslydecreased (P 〈 0. 05,P 〈 0.01) , and the FVC and PEF of TPT treated group were obviously decreased (P 〈 0. 05 ) in comparison with those of normal group. The FVC, FEV1, FEF25, FEF50, MMEF and PEF of XFC treated group were obviously increased in comparison with those of model group ( P 〈 0. 05, P 〈 0. O1 ). The FVC, FEV1, FEF25, FEF50, MMEF and PEF of MTX treated group and the FVC, FEFso and MMEF of XFC treated group were obviously decreased in comparison with those of XFC treated group ( P 〈 0. 05 ,P 〈 0. 01 ). Compared with that of normal group, tumor necrosis factor-α (TNFα) in blood serum decreased obviously in model group (P 〈 0. 05), but interleukin-10 (IL-10) lowered obviously (P 〈 0.01 ). TNFα of XFC treated group was lower than that of model group (P 〈 0. 01 ) , and IL-10 of XFC treated group was higher than that of model group (P 〈 0. 01 ), but there was no obvious difference
