摘要
目的研究威麦宁体外对人高转移肺癌细胞(PG)与血管内皮细胞(HUVEC)黏附作用的影响,探讨其抗肿瘤转移作用机制。方法MTT法检测威麦宁对PG、HUVEC活力的影响;虎红染色法检测威麦宁对PG细胞与HUVEC黏附作用的影响,检测对肿瘤坏死因子(TNF-α)活化的HU-VEC与PG细胞黏附作用的影响。结果威麦宁(31.25、62.5 mg/L)作用PG、HUVEC 24 h后,对两种细胞的存活率即活力皆没有影响(P>0.05)。威麦宁(31.25、62.5mg/L)作用PG细胞24 h后,对HUVEC的黏附有明显抑制作用(P<0.05,P<0.01);作用HUVEC 6、12 h后,能明显降低TNF-α诱导的HUVEC对PG细胞的黏附作用(P<0.01)。结论威麦宁可能通过对PG细胞和HUVEC双重作用,抑制PG-HUVEC间的黏附,从而抑制肿瘤细胞的血道转移。
Objective To study the effect of Weimaining (WMN) on adhesion between high metastasis lung cancer cell line (PG) and human umbilical vein endothelial cell(HUVEC) in vitro to investigate the mechanism of anti -tumor metastasis of WMN. Methods The effects of WMN on the vitality of PG and HUVEC were detected by MTT. The effect of WMN on adhesion of PG to HUVEC and on adhesion of TNF - activated HUVEC to PG were investigated by Rose Bengal staining. Results WMN (31.25,62.5 mg/L) had no effect on PG and HUVEC vitality after treating cells for 24 h( P 〉 0.05 ). WMN (31.25, 62.5 mg/L)obviously decreased PG-HUVEC adhesion after treating PG for 24 h( P 〈 0.05 ,P 〈 0.01 ) HUVEC-PG adhesion decreased significantly after TNF- induced HUVEC was treated by WMN for 6 h and 12 h(P 〈.0.01 ). Conclusion WMN may inhibit the adhesion between PG and HUVEC by effect on both PG and HUVEC. Thereby, WMN can inhibit blood route metastasis of tumor cells.
