摘要
目的:研究survivin在二烯丙基二硫(diallyl disulfide,DADS)诱导HepG2细胞凋亡中的作用。方法:MTT法检测细胞的生长活性;流式细胞仪检测细胞周期;RT-PCR法检测survivin mRNA水平;Western blot法检测survivin蛋白水平。结果:用药组HepG2细胞活性与正常组相比,随着药物浓度的增加(25,50,100,200μmol/l),分别下降了9.3%、10.4%、21.6%、31.2%,HepG2细胞凋亡率分别增加0.83%、1.97%、6.0%、9.9%,低浓度(25,50μmol/l)的DADS可以诱导survivin mRNA和蛋白表达升高,而高浓度的(100,200μmol/l)DADS可以降低survivin mRNA和蛋白表达。结论:DADS诱导HepG2细胞survivin表达增加,抵抗DADS诱导HepG2细胞的凋亡作用。
Objective: To investigate the expression of survivin induced by diaUyl disulfide (DADS), and determine the role in chemotherapy of hepatoceLlular carcinoma (HepG2). Methods: The growth inhibition of HepG2 cells was measured by MTT assay. The Phase distribution of cell cycle and apoptosis was analyzed by flow cytometry. The expression of survivin was detected by RT-PCR and western-blot. Results: After treatment with 25-200μ mol/l DADS for 24 h, MTF assay showed that HepG2 cell viability, compared with the control group, dropped to 9.3%, 10.4%, 21.6% and 31.2%, respectively. Flow cytometry analysis showed that apoptosis rate, compared with the control group, increased 0.83%, 1.97%, 6.0% and 9.9% respectively. RT- PCR assay revealed that DADS of low concentration (25, 50 μmol/l) could induce the increasing expression of survivin mRNA, while the DADS of high concentration ( 100, 200 μmol/1) was opposite. Western blot analysis showed the protein of survivin was decreased with the concentration increase. Conclusion: The expression of survivin was induced in HepG2 by DADS and resisted to DADS-mediated apoptosis.
