摘要
目的检测膀胱移行细胞癌(bladder transitional cell carcinoma,BTCC)中凋亡抑制蛋白sur-vivin、caspase-3的表达,探讨其在膀胱移行细胞癌发生、发展中的意义。方法采用免疫组织化学SP法对60例膀胱移行细胞癌组织和10例正常膀胱黏膜组织中的survivin、caspase-3进行检测。结果膀胱移行细胞癌组织中survivin的蛋白表达率(63.3%)明显高于正常膀胱黏膜组织(20.0%),并随着临床分期的增加,阳性率增加。而caspase-3在膀胱移行细胞癌组织中的阳性率(53.3%)明显低于正常膀胱黏膜组织(90.0%)。survivin与caspase-3的表达呈负相关(r=-0.914,P<0.05)。结论survivin的蛋白高表达和caspase-3的蛋白低表达在膀胱癌的发生和细胞的凋亡调控方面可能起着重要作用。
Objective To investigate the expressions of survivin and caspase - 3 and evaluate their effects on the tumorigenesis and progression in bladder transitional carcinoma( BTCC). Methods The immunohistochemistry ( SP method) was used to determine the expression of survivin and caspase - 3 in 60 cases of BTCC and 10 normal bladder mucosas. Results The protein expression levels of survivin (63.3 % ) in BTCC was significantly higher than those in normal bladder samples(20.0% ), and the levels was correlated with grades of BTCC. But the expression of caspaso - 3 in BTCC (53.3% ) was significantly lower than those in normal bladder samples (90% ) , and its expression level was also related to the grade of BTCC. A significant correlation between survivin and caspase- 3 gene expression in bladder transitional cell cancer was found. There is negative relationship between survivin and caspaso - 3 expression in bladder transitional cell cancer. Conclusion Both over expression of survivin and descended expression of caspaso - 3 may play an important role in the tumorigenesis and apoptotic regulation of bladder tissues.
