摘要
目的建立移植脂肪颗粒活性检测的可靠方法。方法获取健康年轻女性行下腹部脂肪抽吸术的脂肪样本200 mL。样本分为处理组和对照组。其中处理组根据不同离心力分为600、800、1000、2000、3000和4000 r/m in组;对照组则为未经离心处理的样本。分别采用MTT法和葡萄糖转移测定法对离心损伤的脂肪颗粒进行活性检测。对检测的数据分别进行统计学分析,比较各组之间差异;同时对两种检测方法所得数据行相关性分析。结果MTT检测表明,随着离心力的增加,各组脂肪活性呈下降趋势。各处理组间比较有17对数据有显著性差异(P<0.05);葡萄糖转移测定也呈现同一趋势,各处理组间比较有19对数据有显著性差异(P<0.05)。相关性分析表明,MTT法和葡萄糖转移法对脂肪受损活性的检测具有良好的相关性(r=0.9870)。结论葡萄糖转移法测定脂肪活性能准确反应脂肪颗粒的活性状态,具有简便、可靠且灵敏度高的优点。
Objective To establish a method to measure the viability of fat granules for autologous transplantation. Methods Samples of fat tissue (200 mL) were harvested from a healthy young woman by liposuction method. The samples were divided into treatment and control groups. The treatment groups were subdivided according to different centrifugal force: 600, 800,1 000, 2 000, 3 000 and 4 000 r/rain, while the samples without treatment were assigned as control. Both MTT assay and glucose transport test were applied to measure the viability of fat granules in different groups. Analyses of variance were preceded between different groups and analysis of correlation was also performed between the two methods of measuring the viability. Results MTT assay indicated that the viability of fat granules decreased with the increase of centrifugal force. When comparisons were made between each groups, 17 pairs of data exhibited significant difference (P 〈0.05). The same trend was found in glucose transport test, and significant difference was observed in 19 pairs of data (P 〈0.05). Analysis of correlation revealed that MTT assay and glucose transport test were well correlated in the measurement of viability of fat granules ( r = 0. 9870). Conclusion The glucose transport test bears great accuracy and sensitivity in measuring the viability of fat granules.
