摘要 目的研究三丁基氯化锡(TBTCL)XCd,鼠胸腺细胞凋亡及Fas蛋白表达的影响,探讨TBTCL的免疫毒性机制。方法ICR种小鼠体重20-25g,雄、雌各半,共40只,随机分为4组,每组10只,即1个对照组和低、中和高3个组,经口灌胃21d,流式细胞仪检测胸腺细胞凋亡状况,免疫组化染色,观察Fas蛋白在胸腺的表达。结果低、中和高剂量组,胸腺细胞的早期凋亡率高于对照组P〈0.05,且有随剂量增加而升高的趋势,Fas蛋白阳性表达率在染毒组亦强于对照组P〈O.05。结论TBTCL可诱导胸腺细胞凋亡,且可导致Fas蛋白表达增强,这可能是其免疫毒性作用机制之一。 Objective Study the effect of tribuhyltin chloride (TBTCL) on thymocyte apoptosis and Fas protein expression in mice, try to find the mechanism of immunotoxicity. Methods 40 ICR mice were divided into four groups: "the control, the low (0.5 mg /kg), the middle (4 mg/kg) and the high (20 mg/kg), intragastric through mouth for 21 d, detected the thymocyte apoptosis by flow cytometry, observed the fas protein expression in thymus by immunohistocbemistry. Results The earlier period apoptosis rate of thymocyte in the experimental group were higher than the control ( P 〈 0.05), and had ascendent tendency with the dose raised, the Fas protein expression in experimental group was stronger than the control (P 〈 0.05 ). Conclusion TBTCL can induce thymocyte apoptosis, and result in strong expression of Fas protein, which may be one of the mechanism of immunotoxicity.
