摘要 目的利用液相色谱/质谱联用仪(LC/MS)分析叔丁基对苯二酚(TBHQ)在大鼠血清中的代谢物,探讨了TBHQ在大鼠体内的代谢途径。方法通过一级质谱分析,对大鼠血清中TBHQ的代谢物进行鉴别;通过多级质谱分析,对大鼠血清中的TBHQ及其代谢物进行鉴定。结果在大鼠血清中检测到TBHQ(3.5%),TBHQ的代谢物包括硫酸型结合物(93.2%)、葡萄糖醛酸型结合物(1.5%)和一种极性不明代谢物(推测为TBHQ二羟基化物的谷氨酸结合物,1.8%)。结论TBHQ在大鼠血清中主要经Ⅱ相反应(与硫酸或葡萄糖醛酸结合)代谢,此外少量TBHQ会发生Ⅰ相反应,推测为氧化反应,生成的二羟基化物与谷氨酸结合。 Objective To investigate the metabolic pathways of tertiary butylhydrequinone (TBHQ) in rots, liquid chromatography/mass spectrometry (LC/MS) was used to analyze TBHQ and its metabolites in rat serum samples. Methods Major metabolites of TBHQ were differentiated from the other components in serum by MSI analysis and then they were identified by MS^n analysis, Results An average of 3.5% TBHQ, 93.2% TBHQ-sulfate, 1.5% TBHQ-glucuronide and 1.8% polar unidentified metabelites suspected to be the glutamic acid conjugate of dihydroxy-TBHQ was recovered in rat serum. Conclusion The major metabolic pathway of TBHQ in rat serum should be phase Ⅱ reaction (biotransformed by glucuronidation or arylsulfatation), although a small quantity of TBHQ would be metabolized by phase Ⅰ reaction that might be oxidation.
