摘要 目的:探讨叶黄素对食管癌细胞凋亡的影响及其作用机制。方法:EC9706细胞在RPMI培养液(含10%小牛血清)中进行常规传代培养。常规条件下培养24h后,用叶黄素处理,同时设溶剂对照组和5-氟尿嘧啶(5-Fu)阳性对照组。采用流式细胞术观察叶黄素对细胞凋亡的影响,并用免疫组织化学方法检测其对凋亡调节蛋白Bax和Bcl-2表达的影响。结果:在实验条件下,100μg.mL-1叶黄素处理细胞96h可促进细胞凋亡。免疫组化分析结果表明,同样条件下,叶黄素可抑制抗凋亡蛋白Bcl-2的表达并促进促凋亡蛋白Bax的表达。结论:叶黄素具有促进EC9706细胞凋亡的作用,此作用可能是通过调节凋亡相关蛋白Bcl-2和Bax而实现的。 Objective: To study the effects of lutein on apoptosis and its mechanism. Method: The cells of human esophageal carcinoma EC9706 were grown in RPMI medium containing 10% bovine serum and were treated with lutein at 100μg · mL^-1 concentration. Flow eytometry was employed to investigate the effects of lutein on cell apoptosis of EO9706 cells. Histochemistry was performed to determine apoptosis-related protein expresion. Result: Flow eytometry analyses revealed that lutein inered EC9706 cell apoptosis ratio when treated with lutein 100 μg · mL^-1 at 96 h. Lutein decreased the expression of Bel-2 protein and increased the expression of Bax protein in EC9706 cells. Conclusion: Lutein could inhibit mitosis and stimulate apoptosis of EC9706 cells. The apoptotic effect may result from the down-regulation of expression of Bcl-2 and up-regulation expression of Bax.
