摘要
目的:研究树突状细胞(dendritic cells,DC)与肿瘤细胞裂解物致敏的树突状细胞(tumor-associated antigen/dendritic cells,TAA/DC)对荷瘤大鼠的免疫治疗效果。方法:建立大鼠卵巢癌皮下移植瘤模型;于成瘤后2周分别用PBS、DC或TAA/DC进行免疫治疗,比较治疗效果。结果:TAA/DC抑制肿瘤细胞的增殖与转移,对荷瘤大鼠治疗后8周,局部肿瘤无明显增长,P=0.355,无转移灶出现;TAA/DC可明显改善大鼠免疫状况,对荷瘤大鼠治疗后3周,使已经下降的CD8+T细胞百分比又明显上升,P=0.000;TAA/DC对荷瘤大鼠单次免疫治疗的效果具有时限性,在对荷瘤大鼠治疗后8周,CD8+T细胞百分比再次出现明显下降,P=0.000。结论:TAA/DC可改善荷瘤大鼠的细胞免疫状态,抑制肿瘤细胞的增殖与转移。
OBJECTIVE: To study the immunotherapy capacity of DC and TAA/DC to tumor burdened rats in vivo. METHODS: The animal model was established which inoculated with ovarian adenocarcinoma cells. Two-week tumor burdened rats were treated immunologically with PBS, DC and TAA/DC respectively, and the results of the treatment were compared. RESULTS: TAA/DC inhibited the hyperplasia of tumor cell, and made subcutaneous tumor retarded,P= 0. 355. In TAA/DC treated tumor carring rats, no metastasis was observed. TAA/DC improved the immune condition of the tumor-burdened rats that CD8^+ T cells raised remarkably, P=0. 000. However,the effect of improved immune condition only sustained a short period, and showed that CD8^+ T cells reduced before the death,P= 0.000. CONCLUSIONS: Specific anti-tumor immunotherapy of TAA/DC could make the immune condition of the tumor earring rat much better. The hyperplasia of tumor cells is inhibited, and the rate of metastasis is reduced.
