脂质体介导的hTERT基因反义核酸对K562细胞端粒酶活性的影响

摘要 目的研究人端粒酶逆转录酶(Human telomerase reverse transcriptase,hTERT)基因反义核酸(anti-sense phosphorothioate oligodeoxynucletide,ASODN)对K562细胞端粒酶活性的影响。方法本研究采用脂质体介导的基因转染方法将ASODN导入K562细胞,以TRAP-ELISA法检测药物对端粒酶活性的影响,免疫荧光检测hTERT蛋白表达水平的变化。结果ASODN能显著抑制K-562细胞端粒酶活性和hTERT基因的表达,呈时间依赖性。结论以hTERT基因ASODN抑制端粒酶活性是治疗白血病的潜在途径。 Objective To investigate the effect of Human telomerase reverse transcriptase (hTERT) gene antisense phosphorothioate oligodeoxynueletide (ASODN) on telomerase activity in K562 cells. Methods By using liposome mediated DNA transfeetion, telomerase antisense RNA was successfully introduced into k562 cells. The telomerase activity of K562 cells was examined by telomerie repeat amplification protocol(TRAP)-enzyme-linked immunosorbent assay(ELISA). The expression levels of hTERT mRNA protein was assayed by immunofluoreseenee using fluorescence isothioeyanate label. Results TRAP- ELISA and immunofluoreseenee assay showed that the telomerase activity and expression levels of hTERT mRNA protein from K562 cells decreased with time after hTERT ASODN treatment. Conclusion It seems to be a new method to treat leukemia by using telomerase antisense to inhibit telomerase activity of K562 cells, telomerase is a new latent approach to treat leukemia.
