17β-雌二醇下调人成骨样MG63细胞结缔组织生长因子表达机制的研究

Investigation on the mechanism of 17β-estradiol-downregulating connective tissue growth factor expression in human osteoblastic MG63 cells

摘要 10μmol/L雌二醇可明显下调人成骨样MG63细胞结缔组织生长因子(CTGF)。RNA及蛋白的表达;蛋白激酶A(PKA)阻断剂H-89可完全阻断雌二醇对CTGF的降调节作用。PKA活化剂forskolin可模拟雌二醇作用,明显下调人成骨样MG63细胞CTGF mRNA、蛋白的表达。17β-雌二醇下调人成骨样MG63细胞CTGF表达由PKA介导。 17βestradiol (10-s tool/L) decreased the expression of CTGF mRNA and protein, which could be completely blocked by H-89, protein kinase A (PKA) antagonist. Forskolin ( PKA agonist) can mimic the effect of estradiol on CTGF expression and reduce the expression of CTGF. 17β-estradiol can inhibit the CTGF expression via PKA pathway in human osteoblastic-like MG63 cells.
