摘要
目的建立反相高效液相色谱法测定血浆中卡维地洛的浓度。方法采用反相高效液相色谱法测定血浆中卡维地洛的浓度，选用Shim-pack VP-ODS柱（150mm×4．6mm，5μm），[甲醇：水（60：40）]-乙腈（88：12）为流动相，流速：1ml／min，检测波长：280nm。结果卡维地洛血药浓度的线性范围为25～2000ng·ml^-1（r=0．9999），提取回收率70．84％，方法回收率102．05％,日内、日间RSD分别为1．91％、5．58％。结论该方法准确、灵敏，适于卡维地洛药代动力学研究。
Aim An RP-HPLC method was developed for detemination of Carvedilol in human plasma. Methods Carvedilol was extracted from plasma and the applied column was Shim-pack VP-ODS （150mm ×4.6mm,5μm）. The mobile phase consisted of methanol and water（60： 40）- acetonitrile（88： 12）. Carvedilol was detected by UV absorption at 280nm. Results The standard curve was liner in the range of 25 - 2 000ng.ml^-1（ r =0.999 9） ;the extracted recovery of Carvedilol was 70. 84% ;the recovery of the method was 102.05% ;and the RSDs of intra-day and inter-day were 1.91% and 5.58%. Conclusion the method is sensitive and accurate.
