琼枝麒麟菜多糖对辐射损伤小鼠脾细胞免疫功能的影响 被引量:7

摘要 目的观察琼枝麒麟菜多糖(EGP)对辐射损伤小鼠脾细胞免疫功能的影响,为临床用于辐射损伤的治疗提供科学依据。方法将50只小鼠随机分为正常对照组(A组)、辐射对照组(B组)和3个不同剂量EGP+辐射组(C、D、E组),C、D、E组分别以3个不同剂量(100、200和400mg.kg-1.d-1)按每10 g体质量0.1 mL灌胃,连续10 d;A组和B组均用等量生理盐水灌胃,7 d后除A组外均行一次性γ射线辐射,每只2 Gy,20 h后检测脾细胞对刀豆蛋白(ConA)及脂多糖(LPS)的增殖反应活性,脾混合淋巴细胞反应(MLR),脾细胞DNA及蛋白质合成。结果D、E组小鼠脾细胞对ConA的增殖值、MLR值、脾细胞DNA及蛋白质合成值(4202±1042、6302±2235;2302±4873、379±1024;2415±531、3827±937;1541±164、2098±536)cpm,均明显高于B组(2401±640;1028±295;868±169;782±307)cpm(P<0.01)。E组小鼠脾细胞对LPS刺激的增殖值(2287±585)cpm明显高于B组(1078±398)cpm(P<0.01)。结论琼枝麒麟菜多糖(EGP)对辐射损伤脾细胞的免疫功能具有一定的调节作用。 Objective To investigate the effect of Eucheuma gelatinae polysaccharide (EGP) on the immune function of splenocytes in γ-rays irradiated mice, so as to provide scientific basis for clinical therapy for irradiated injury. Methods Fifty mice were randomly divided into 5 groups, normal control group (A group), irradiated control group (B group) and 3 EGP treatment groups(C,D,E groups). C,D,E groups were fed with EGP 100,200,400 mg· kg^-1·d^-1 respectively for 10 days, and A, B groups were fed with equal volume of physiological saline. On the 7th d, B, C, D, E groups were irradiated with γ-rays (2.0 Gy). Proliferation response of splenocytes to ConA and LPS,mixed lymphocytes response(MLR), DNA and protein synthesis of splenocytes were examined 20 h after irradiation. Results The proliferation response of splenocytes to ConA, MLR, DNA and protein synthesis of splenocytes in D, E groups treated with EGP (4202 ± 1042,6302 ± 2235;2302 ± 487, 3379 ± 1024; 2415 ± 531, 3827 ± 937; 1541 ± 164, 2098 ± 536) cpm respectively] were significantly greater than those in B group[(2401 ± 640, 1028 ± 295, 868 ± 169,782± 307)cpm] ( P 〈 0. 01 ), and proliferation response of splenocytes to LPS[ (2287 ± 585)cpm] in E group was greater than that in B group ( 1078 ± 398)cpm] ( P 〈 0.01). Conclusion Eucheuma gelatinae polysaccharide has some modulatory effect on the immune function of splenocytes injured by irradiation.
