摘要
为进一步探讨内皮素参与烧伤后病理生理学改变的机制,分离和培养大鼠心脏微血管内皮细胞。分别用正常大鼠和烧伤大鼠(30%TBSAⅢ度)伤后6小时的血清培养心脏微血管内皮细胞12小时,用放射性配基结合受体分析法,观察烧伤血清对心脏微血管内皮细胞膜内皮素受体亲和力及结合容量的影响。结果表明,大鼠心脏微血管内皮细胞膜上只有内皮素受体亚型B分布,该受体的最大结合容量(Bmax)为142±27fmol/mg蛋白,平衡解离常数(Kd)为119±22pmol/L。烧伤血清不影响该受体的Kd值,但使Bmax增加68%,表明烧伤血清上调内皮素受体亚型B。提示烧伤后微血管内皮细胞对内皮素作用的敏感性增加。
Cardiac microvascular endothelial cells(CMEC) were harvested and cultured. Following incubation of CMEC with normal or burn rat (30%TBSA, Ⅲ°) serum for 12 hours, respectively, the affinity and binding capacity of endothelin(ET) receptor in CMEC membranes were investigated by radi- oligand binding assay. The results showed that only ET_B receptor was distributed in rat CMEC mem- branes, with a maximal binding capacity(B_(max)) of 142±27 fmol/mg protein and a dissociation constant (Kd) of 119±22 pmol/L. Burn rat serum induced an increase in B_(max) of ET_B receptor by 68%, without changing its Kd value,demonstrating that burn rat serum up-regulated ET_B receptor. These results sug- gest that responsiveness to ET of microvaseular endothelial cells may increase following burn.
