摘要
目的:探讨针刺抗脑缺血再灌注炎性损伤的机制。方法:采用线栓法制作大鼠大脑中动脉缺血模型(MCAO),采用免疫组织化学技术研究P-选择素和E-选择表达的动态变化。结果:再灌注3h各实验组P-选择素和E-选择素均开始表达,于再灌注24h均达到高峰(组内比较P<0.01),持续至再灌注48h,针刺可显著降低P-选择素和E-选择素的表达(与模型组和非穴组比较,P<0.01)。结论:P-选择素和E-选择素的表达上调与再灌注后的炎症反应有关,早期针刺治疗可能通过下调脑缺血区粘附分子P-选择素和E-选择素的表达而防治脑缺血再灌注炎性损伤。
Objective: To study mechanisms of early treatment on cerebral ischemia-reperfusion (I/R) by acupuncture in resisting inflammation injury in rats. Methods: Cerebral I/R Model was established occlusion of the middle cerebral artery for lhr and then remoral of the occlusion for 3hr, 12hr, 24hr and 48hr respectively.The DNA expression of P-seleetin and E-selectin was displayed by using Imanohistochemieal ABC in ischemie cerebral region in rats. Result: The DNA expression of P-seleetin and E-seleetin was detected at capillary walls in postiehemia cerebral cortex at lhr ischemia-3hr reperfusion and peaked at lhr isehemia-24hr reperfusion and increased continually during 48hr reperfusion. The DNA expression of both P-selectin and E-seleetin in ischemie cerebral cortex were downregulated considerably by acupuncture treatment ( P 〈 0.01 ) .Conclusion: The protective effect of acupuncture on cerebral neurons under acute infarction is pussiblely through its downregulative action on the expression of P-seleetin and E-selectin, which caused neutrophil infiltration induced by adhesive molecules between endothelium cell and neutrophil.
