摘要
抑制消减杂交(Suppressionsubtractivehybridization,SSH)是以消减杂交为基础结合抑制PCR方法发展起来的分离克隆差异表达基因的一种策略。该方法具有简便易行、特异性高、背景低、重复性好、能分离出丰度较低的特异性片段等特点。主要介绍其原理、方法及其应用,并对方法的优缺点和应用前景作分析。
A highly effective method, termed suppression subtractive hybridization (SSH), has been developed for the generation of subtracted cDNA libraries of differential expressed genes. It is based primarily on a technique called suppression PCR and combines normalization and subtraction in a single procedure. It has many advantages such as easy to manipulate, with high specificity, lower background, better repeatability and easy to separate low abundance fragments. This article mainly focus on the principles and it' s application in various research fields.
