摘要
目的将携带特异性抗体的杂交瘤细胞作为天然抗体＿载体，进行血清学相关抗原检测的方法学研究。方法运用特异性杂交瘤细胞ＥＬＩＳＡ双抗体夹心法，测定一组消化道癌标本包括胃癌６９例，其他消化道癌６３例，胃良性病３５例，以及健康体检者８４例。结果正常值判定为００７６±００７６（ｘ±２ｓ），特异性阻断试验为７３％，回收率为６８４％，批内ＣＶ值为５２％－８９％，批间ＣＶ值为１１７％－１７６％。结论特异性杂交瘤细胞ＥＬＩＳＡ方法测定血清中肿瘤相关抗原，方法简单，具有较强的特异性及敏感性，可应用于临床。
AIMS Using specific hybridoma cell as natural antibody carrier, we established a SHC_ELISA in detection of serum gastric cancer. METHODS A series of the malignant tumor sera of digestive tract gastric cancer, 69; other digestive tract cancer, 63; benign gastric diseases, 35; and healthy subjects, 84 were detected by SHC_ELISA. RESULTS The range of normal value was 0 076%-0 076%(±2s), specific inhibition rate was 73%, recovery rate 68 4%,within run and between run CVS were 5 2% to 8 9% and 11 7% to 17 6%. CONCLUSIONS SHC_ELISA is simpl, specific, sensitive, and may be used chinically.
