乌司他丁对缺氧缺血性新生鼠的脑保护作用 被引量:5

The protective effect of ulinastatin on newborn rats with hypoxic-ischemic brain damage

摘要 目的探讨乌司他丁对新生鼠缺氧缺血性脑损伤(HIBD)的作用。方法选7日龄新生Wistar大鼠60只制备HIBD模型。乌司他丁治疗组大鼠分别于术后即刻、术后8、16h腹腔注射乌司他丁一次,术后24h处死。取脑组织迅速秤重,固定,切片行HE和TUNEL染色,测定凋亡细胞数,对皮层脑细胞变性坏死和胶质细胞增生行病理量化评分,电镜观察海马区细胞形态。结果治疗组在脑重增加值及左、右脑重差值、凋亡细胞数和病理量化评分明显低于缺氧缺血组(P<0·01);各治疗组之间比较上述指标无统计学意义(P>0·05)。结论乌司他丁可减轻缺氧缺血后脑细胞水肿、变性、坏死及凋亡,对新生鼠HIBD具有保护作用。 Objective To study the effect of Ulinastatin on newborn rats with hypoxic-ischemic brain damage(HIBD) Methods Sixty newborn Wistar rats aged 7 days old were selected to establish HIBD model. In therapeutic group. Ulinastatin was injected peritoneally at instantly, 8 hours and 16 hours after procedures, then the rats were sacrificed 24 hours after hypoxic and ischemic insults. Their brain tissues were weighted and fixed, the slices were stained by HE and TUNEL. The number of cells with apoptosis was counted. The degeneration, necrosis and hyperplasia of gliocytes in cerebral cortex cells were measured with quantitative pathological score. The cells of hippocampus were examined by electron-microscopy. The results were compared among HIBD model group, sham operation group, control group and three Ulinastatin treated groups. Results The values of brain weight increased, the weight differences of left and right brain, the number of apoptotic cells and quantitative pathological score of brain tissue in therapeutic group were much lower than that of HIBD group(P 〈0.01 ). There were no significant differences of above values among all three therapeutic groups ( P 〉 0. 05 ). Conclusions Uhnastatin treatment can decrease cerebral cellular edema, degeneration, necrosis and apoptosis after hypoxic-ischemic insult, and it provides protective effect on brain tissues.
