褐藻多糖GS201对鼠脊髓损伤后神经细胞的保护及功能恢复作用 被引量:1

摘要 目的:探讨褐藻多糖GS201对大鼠脊髓损伤后神经细胞的保护及功能恢复的作用。方法:75只Wistar雄性大鼠,随机分为假手术组(A组)、对照组(B组)和治疗组(C组),每组25只,A组只打开椎板,不损伤脊髓,B组脊髓损伤后腹腔注射等渗盐水,C组脊髓损伤后1h腹腔注射褐藻多糖GS20110mg/kg,此后每天1次,持续治疗5d。末次治疗后每组处死5只动物,检测脊髓组织超氧化物歧化酶(SOD)活性,丙二醛(MDA)和一氧化氮(NO)含量,脊髓损伤后第1小时和末次治疗后第1、5、10天评定后肢运动功能(Salzman评分)和检测运动诱发电位(MEP)。结果:SOD活性C组高于B组(P<0.01),MDA和NO含量C组低于B组(P<0.01)。脊髓损伤后,后肢运动障碍,MEP波幅(P1-N1峰峰值)降低,潜伏期延长,末次治疗后各时间内C组MEP波幅、潜伏期、神经功能恢复均优于B组(P<0.01或P<0.01)。结论:褐藻多糖GS201能减少脊髓损伤后氧自由基所致继发损伤,并促进神经功能恢复。 Objective To explore the effects of sulfated polysaccharides from brown seaweeds GS201 on the protection and function recovery of neurons after spinal cord injury in rats. Methods 75 Wistar male rats were randomly assigned to sham-operated group (group A), control group (group B), and GS201 treatment group (group C). The rats in group A received lamina of vertebral opening with intact spinal cord; The rats in group B received intraperitoneal injection of NS after the impairment of spinal cord while the rats in group C received sulfated polysaccharides from brown seaweeds GS201 (10 mg/kg) daily for 5 consecutive days. Five of them in each group were executed after the end of the treatment. The SOD activity in the spinal tissue and the levels of MDA and NO were detected. The motor function of the posterior limbs was evaluated with Salzman's score and MEP was measured at lh after spinal cord injury, and on days 1, 5 and 10 following the end of the therapy. Results The SOD activity in group C was higher than that in group B (P 〈 0.01) while the levels of MDA and NO were lower (P 〈 0.01). After spinal cord was impaired, dyskinesia of the lower limbs occurred and the peak values of P, to N, on MEP amplitudes were decreased with a prolong latent period. The normalization of the MEP amplitudes and the latent period, and the function recovery of the impaired nerve in group C were superior to those in group B on days 1, 5 and 10 after the end of the treatment (P 〈 0.05 or P 〈 0.01 ). Conclusion Sulfated polysaccharides from brown seaweeds GS201 can relieve the secondary damage induced by oxygen free radicals after spinal cord injury and be beneficial to the function recovery of the impaired nerve.
