摘要
目的:探讨脑络欣通对气虚血瘀证脑缺血再灌注大鼠神经元、神经胶质细胞形态学变化和TNF -α、c-Myc蛋白表达的影响。方法:采用气虚血瘀证模型鼠,再线栓大脑中动脉复制局部脑缺血再灌注模型,缺血2小时分别再灌注1天,3天,7天,随机分为假手术组、模型组、益气组、活血组和脑络欣通组。应用光镜和免疫组化法,观察缺血侧额顶叶皮质神经元、神经胶质细胞形态学变化及TNF-α、c-Myc蛋白表达。结果:气虚血瘀证局灶性脑缺血再灌注后,同侧额顶叶皮质梗死区神经元、小胶质细胞、星形胶质细胞出现变性、死亡,梗死灶周围小胶质细胞和星形胶质细胞增生。TNF-α、c-Myc蛋白表达阳性细胞(positive cells)和平均吸光度(A value)增加,与假手术组比较均有显著性差异(p<0.01)。脑络欣通可明显改善大鼠神经元和神经胶质细胞的形态变化,能使TNF-α、c-Myc蛋白表达降低,与模型鼠比较有显著性差异。益气组、活血组部分时段表达降低,脑络欣通组降低明显,与益气组、活血组比较部分有显著差异。结论:脑络欣通及其拆方可通过抑制TNF-α、c-Myc蛋白的表达来调节不同细胞间相互作用,进而影响气虚血瘀证大鼠脑缺血再灌注后神经元、神经胶质细胞之间在损伤、抗损伤及修复中的相互作用,减轻病理性损伤。
Objective:To investigate the effects of naoluo xintong on morphologic changes in neuron or neuroglial cells and TNF-α,c-Myc protein ex- pression in the cortex of frontal or parietal lobe following local cerebral ischemia-reperfusion in MCAO Rats.Methods:The model was esta- bished by intraluminal thread occlusion of the middle cerebral arteries(MCAO) after qi-deficiency and blood stasise syndrome model was es- tablished by high lipid diet,hunger and swiming.The morphologic changes in neuron or neuroglial cells and expression of TNF-α,c-Myc protein were observed by techniques of immuno-histochemical staining and optical microscopy. Results:The degeneration or necrosis of neuron or neuroglial cells were observed at the center of infarction,it was very serious at 3d after reperfusion.Astrocyte and microglial cells proliferation were observed in the broder of infarction,it was very obvious at 7d after reperfusion.The value and positive cells amount of TNF-αc-Myc protein expression in model group were higher than those in sham,those in naoluo xintong group were lower than those in model group.Conclusion:Naoluo xintong could regulate inreacfion between neuron and neuroglial cells,allevi- ate pathologic injury after focal cerebral ischemia-reperfusion in MCAO rats by inhibiting the expression of TNF-α,c-Mye protein.
