摘要
目的观察大鼠骨髓间充质干细胞(MSCs)体外培养、定向诱导分化为心肌样细胞的条件。方法抽取大鼠骨髓,采用贴壁培养法分离MSCs进行培养、传代,在第4代MSCs中添加诱导剂5-氮胞苷(5-aza)对其进行诱导分化,观察细胞形态学的变化,应用荧光免疫组织化学方法进行鉴定。结果大鼠MSCs贴壁呈集落生长,有成纤维细胞样外观。诱导后细胞呈梭形,排列方向渐趋一致,有肌管样结构形成,肌钙蛋白及Connexin43表达强阳性。结论大鼠MSCs体外在5-氮胞苷作用下可定向分化为心肌样细胞。
Objective To investigate the culture and myocardial differentiation of rat bone mesenchymal stem cells(BMSCs) in vitro. Methods MSCs were isolated from rat bone marrow using anchoring culture,5-azacytidine (5-aza) for myocardial induction was added to the culture medium, the cell morphology was noticed, Differentiation was identified by immunochemistry technique. Results The MSCs were found grew into colonies with the appearance of fibroblastic cells, MSCs morphology were spindle - shaped after induction ,The direction of the cells arraying was similar gradually, Passage4(P4) MSCs showed formation of myotube and expressed cardiomyocyte-associated markers ,Troponin T and Connexin43. Conclusions. MSCs could be induced to differentiate into cardiomvocvte-like cells by 5-aza.
