感染周期型马来丝虫的中华按蚊酯酶同工酶及可溶性蛋白质含量的分析研究 被引量:1

摘要 应用聚丙烯酰胺圆盘电泳(PAGE)和薄层扫描仪对感染周期型马来丝虫的中华按蚊与未感染蚊的酯酶(EST)同工酶进行了酶谱区带、酶活力的分析与比较,并测定了两组蚊体的可溶性蛋白质含量。结果表明,感染蚊与未感染蚊相比,感染蚊的EST同工酶区带总数、主带数、扫描吸收峰及其主峰数均多,且EST同工酶13、15号主带宽、色深,EST酶活力高。感染蚊独具1~2条次带,在扫描图上可见相应的吸收峰。感染蚊平均每只蚊的可溶性蛋白质含量低于未感染蚊。此结果为从生化方面寻求蚊媒监测丝虫病流行的指标提供了线索和依据。 In this paper,the esterase(EST) isozymes of adult female Anopheles Sinensis at various times after taking noninfected and Peridic Brugia malayi-infected blood meal were compared and analysed by disc electrophoresis in polyacrylamide gel and CAMAG TLC SCANNER. The Soluble protein contents in two groups of mosquitoes were tested by Hartree's Method. The result shows that numbers of EST isozyme total bands, prominent bands, total peaks and prominent peaks in the TLC tracings of infected mosquitoes in 3d, 6d, 9d after taking blood meal were more than those of noninfected ones. Prominent band 13 and 15 infected mosquitoes were wider and stained stronger than those in noninfected ones during each life stage. There were γ-2 special EST bands with corresponding scanned peaks in infected mosquitoes. The EST a ctivity was estimated by zymogram scanned area. Either average E activityin an infected adult female mosquito or relative EST activity was higher than that of noninfected one. The average amounts of soluble protein in infected mosquitoes were lower than those of noninfected ones duritig the same life stage. The above results may provide clues and basis to seak targets monitoring mosquito medium of filariasis in the field of biochemistry.
