摘要
目的:克隆新的与激活素受体ⅡA(ActRⅡA)相互作用蛋白,即激活素受体相互作用蛋白4(ActRIP4)基因。方法:应用ActRIP2基因片段作为探针,从小鼠cDNA文库中钓取ActRIP4 cDNA;采用哺乳细胞双杂交进一步确定ActRIP4与ActRⅡA的相互作用;RT-PCR检测ActRIP4 mRNA在组织中的表达情况。结果:克隆的ActRIP4基因全长691bp,ORF区编码118个氨基酸残基;ActRIP4与ActRⅡA具有特异性结合作用;天然的ActRIP4 mRNA在小鼠多种组织中表达。结论:ActRIP4属于ActRIP家族新成员,可以与ActRⅡA相互作用,可能在多种组织中介导激活素受体信号传导过程。
Objective To clone the gene of a novel aetivin receptor-interacting protein 4 (AetRIP4) which interacts with type Ⅱaetivin receptor (AetRⅡ A) . Methods The ActRIP4 gene from a mouse eDNA library was cloned by using the AetRIP2 gene fragment as the probe. By mammalian cell two-hybrid, its ability to combine with AetR Ⅱ A was confirmed. The expressions of AetRIP4 mRNA in tissues were analyzed by RT-PCR. Results The cloned fulllength eDNA for AetRIP4 was 691bp, which ORF was encoding 118 amino acids. The specific mammalian cell two- hybrid analysis revealed that AetRIP4 could specifically combine with AetR Ⅱ A. The results of RT-PCR showed that ActRIP4 mRNA expressed in many tissues. Conclusion AetRIP4 is a novel member of ActRIP family, and has the ability to specifically combine with ActR ⅡA, which may mediate signaling transduction of activin receptor in various tissues.
